Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.

Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
Rhodesians made up an integral component of the Long Range Desert Group (LRDG), a mechanised reconnaissance and raiding unit formed in North Africa in 1940 to operate behind enemy lines. Initially made up of New Zealanders, the unit's first British and Rhodesian members joined in November 1940. It was reorganised several times over the next year as it expanded and by the end of 1941 there were two Rhodesian patrols: S1 and S2 Patrols, B Squadron. Each vehicle bore a Rhodesian place-name starting with "S" on the bonnet, such as "Salisbury" or "Sabi". From April 1941 the LRDG was based at Kufra in south-eastern Libya. The Rhodesians were posted to Bir Harash, about 160 kilometres (99 mi) to the north-east of Kufra, to patrol, hold the Zighen Gap and guard against a possible Axis attack from the north. For the next four months they lived in near-total isolation from the outside world, an exception coming in July 1941 when they and a group of airmen from No. 237 Squadron celebrated Rhodes Day together in the middle of the Cyrenaican desert. In November 1941 the British Eighth Army, commanded by General Cunningham, launched Operation Crusader in an attempt to relieve Tobruk. The British XXX Corps, led by the 7th Armoured Division ("the Desert Rats") with its Rhodesian platoons, would form the main body of attack, advancing west from Mersa Matruh, then sweeping around in a north-westerly direction towards Tobruk. The XIII Corps would concurrently advance north-west and cut off Axis forces on the coast at Sollum and Bardia.
== Diagnosis == No specific test exists to diagnose polymyalgia rheumatica; many other diseases can cause inflammation and pain in muscles, but a few tests can help narrow down the cause of the pain. Limitation in shoulder motion or swelling of the joints in the wrists or hands, are noted by the doctor. One blood test usually performed is the erythrocyte sedimentation rate (ESR) which measures how fast the patient's red blood cells settle in a test tube. The faster the red blood cells settle, the higher the ESR value (measured in mm/hour), which suggests that inflammation may be present. Many conditions can cause an elevated ESR, so this test alone is not proof that a person has polymyalgia rheumatica. Another test that checks the level of C-reactive protein (CRP) in the blood may also be conducted. CRP is produced by the liver in response to an injury or infection, and people with polymyalgia rheumatica usually have high levels. However, like the ESR, this test is also not very specific. Polymyalgia rheumatica is sometimes associated with temporal arteritis, a condition requiring more aggressive therapy. To test for this additional disorder, a biopsy sample may be taken from the temporal artery.
=== Coelomata hypothesis === Before Aguinaldo's Ecdysozoa proposal, one of the prevailing theories for the evolution of the bilateral animals was based on the morphology of their body cavities. There were three types, or grades of organization: the Acoelomata (no coelom), the Pseudocoelomata (partial coelom), and the Eucoelomata (true coelom). Adoutte and coworkers were among the first to strongly support the Ecdysozoa. With the introduction of molecular phylogenetics, the coelomate hypothesis was abandoned, although some molecular, phylogenetic support for the Coelomata continued until as late as 2005.
Smoking has become less popular, but is still a large public health problem globally. Worldwide, smoking rates fell from 41% in 1980 to 31% in 2012, although the actual number of smokers increased because of population growth. In 2017, 5.4 trillion cigarettes were produced globally, and were smoked by almost 1 billion people. Smoking rates have leveled off or declined in most countries, but are increasing in some low- and middle-income countries. The significant reductions in smoking rates in the United States, United Kingdom, Australia, Brazil, and other countries that implemented strong tobacco control programs have been offset by increasing consumption in low income countries, especially China. The Chinese market now consumes more cigarettes than all other low- and middle-income countries combined. Other regions are increasingly playing larger roles in the growing global smoking epidemic. The WHO Eastern Mediterranean Region (EMRO) now has the highest growth rate in the cigarette market, with more than a one-third increase in cigarette consumption since 2000. Due to its recent dynamic economic development and continued population growth, Africa presents the greatest risk in terms of future growth in tobacco use. Within countries, patterns of cigarette consumption also can vary widely. For example, in many of the countries where few women smoke, smoking rates are often high in males (e.g., in Asia). By contrast, in most developed countries, female smoking rates are typically only a few percentage points below those of males.
Retinal pigmented epithelial cells (RPE) and retina have among the highest glyoxalase activities in the body, however, glyoxalase activity is depressed upon aging. This is consistent with observed increases in AGEs associated with aging. Enhancing the glyoxalase system has been shown to delay accumulation of AGEs and associated retinal damage in animals that consume higher glycemic index diets.

Every polyhedron has a dihedral angle at every edge describing the relationship of the two faces that share that edge. This dihedral angle, also called the face angle, is measured as the internal angle with respect to the polyhedron. An angle of 0° means the face normal vectors are antiparallel and the faces overlap each other, which implies that it is part of a degenerate polyhedron. An angle of 180° means the faces are parallel, as in a tiling. An angle greater than 180° exists on concave portions of a polyhedron. Every dihedral angle in a polyhedron that is isotoxal and/or isohedral has the same value. This includes the 5 Platonic solids, the 13 Catalan solids, the 4 Kepler–Poinsot polyhedra, the 2 convex quasiregular polyhedra, and the 2 infinite families of bipyramids and trapezohedra.
=== Gingival crevicular fluid as a biomarker source === For research purposes, utilisation of GCF (gingival crevicular fluid) or PISF (peri-implant sulcular fluid) have been proven to be reliable indicators to examine gingival health, though this is rarely done in clinical practice. Through saliva samples obtained from individuals, the GCF/PISF mixed within saliva have been studied for the level of cytokines and inflammatory mediators. Examples of these mediators include Interleukin (IL) 1-ß, IL-6, macrophage chemotactic protein, interferon g-induced protein 10 (IP-10) and vascular endothelial growth factor (VEGF). Results indicate that individuals with periodontal disease have a significant increase in cytokine count compared to individuals who are of good gingival health. This is explained to be due to the increased interaction between the bacterial biofilm and the cells of the periodontal tissue, leading to heightened diffusion of these mediators along with the GCF into the gingival sulcus.
On September 13, 2011, the Department of the Interior strongly urged the Cherokee Nation to restore voting rights and benefits to descendants of Cherokee Freedmen, including the right to vote in the special election for Principal Chief, at the risk of violating its constitution and the US Constitution. On September 14, the Cherokee AG recommended reinstatement of the Freedmen, pending a hearing for oral arguments. On September 20, Judge Henry H. Kennedy Jr. of the US District Court announced the Cherokee Nation, Freedmen plaintiffs and US government had come to an agreement in a preliminary hearing to allow the Freedmen to vote, with voting to continue through October 5 if necessary. On August 30, 2017, the United States District Court for the District of Columbia ruled in favor of the Freedmen descendants and the U.S. Department of the Interior, granting the Freedmen descendants full rights to citizenship in the Cherokee Nation. The Cherokee Nation has accepted this decision, effectively ending the dispute. In 2021, Shawna Baker, a justice on the Cherokee Nation Supreme Court, published the written opinion, Effect of Cherokee Nation v. Nash & Vann v. Zinke, CNSC-2017-07.
=== Treatment after exposure === The primary treatment is removal of the offending substance. Capsaicin is soluble in alcohol, which can be used to clean contaminated items. Plain water is ineffective at removing capsaicin in most situations, but flushing continuously with water may be used when no alternative is available, such as when treating the eyes or skin. When capsaicin is ingested, cold milk may be an effective way to relieve the burning sensation due to caseins in milk, and the water of milk acts as a surfactant, allowing the capsaicin to form an emulsion with it.

=== 6 November === The RSF agreed to a three-month humanitarian truce agreement proposed by the Quad countries (the United States, Saudi Arabia, the UAE, and Egypt). Meanwhile, the SAF rejected the humanitarian truce, citing crimes against civilians in El Fasher, as well as saying it is a ploy to allow the RSF consolidate control over Darfur and parts of Kordofan.
In atherosclerosis, new blood vessels form within plaques, contributing to their growth and instability. These vessels are often fragile, allowing inflammatory cells and fats to enter, which can cause bleeding inside the plaque and increase the risk of rupture. Some studies in animal models suggest that blocking this vessel growth can reduce atherosclerotic progression. In a myocardial infarction, blocked blood flow deprives heart tissue of oxygen, leading to cell damage. Neovascularization in the surrounding area can help restore oxygen supply and limit further injury. Growth factors such as basic fibroblast growth factor (bFGF) and brain natriuretic peptide (BNP) can promote angiogenesis after an acute MI. Following a stroke, ischemic penumbra (the region surrounding the infarct core) can disrupt the cerebral blood flow. Post-stroke angiogenesis in the area helps restore perfusion and supports neurological recovery. Additionally, arteriogenesis contributes to post-stroke blood flow restoration. Various immune cells and cytokines play a role in regulating angiogenesis after ischemic injury.
In December 2012, the U.S. state of Washington became the first state to officially legalize cannabis in a state law (Washington Initiative 502) (but still illegal by federal law), with the state of Colorado following close behind (Colorado Amendment 64). On 1 January 2013, the first cannabis "club" for private marijuana smoking (no buying or selling, however) was allowed for the first time in Colorado. The California Supreme Court decided in May 2013 that local governments can ban medical cannabis dispensaries despite a state law in California that permits the use of cannabis for medical purposes. At least 180 cities across California have enacted bans in recent years. On 30 April 2024, the United States Department of Justice announced it would move to reclassify cannabis from a Schedule I controlled substance to the somewhat less restricted Schedule III. On 18 December 2025, Donald Trump signed an executive order to reclassify cannabis as a Schedule III drug. In December 2013, Uruguay became the first country to legalize growing, sale and use of cannabis. After a long delay in implementing the retail component of the law, in 2017 sixteen pharmacies were authorized to sell cannabis commercially. On 19 June 2018, the Canadian Senate passed a bill and Prime Minister Justin Trudeau announced the effective legalization date as 17 October 2018. Canada is the second country to legalize the drug. In November 2015, Uttarakhand became the first state of India to legalize the cultivation of hemp for industrial purposes.
== Assessment == Due to Alström Syndrome's effect on nearly every organ system of the body, experts strongly recommend a multidisciplinary team of professionals with experience caring for those with AS when possible. The following multidisciplinary assessment is recommended for those with AS:
== Case study == In 1988, Summons et al. studied the Proterozoic Kwagunt Formation of the Chuar Group in Grand Canyon, Arizona. This sedimentary rock is 850 million years old. After performing an extraction of the rocks with organic solvents, Summons characterized the abundance of various lipid biomarkers using GC-MS/MS, as described above. Using the 412-->191 m/z transition, they identified gammacerane in the extract. Summons interpreted this signal as the diagenetic product of tetrahymanol. At the time, this lipid had only been observed in protozoa, mainly ciliates. They interpreted it as a biomarker for the existence of protozoa in the Neoproterozoic. This report is still the oldest observation of gammacerane in the rock record.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
