Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.

The official language is Romanian, a Romance language (the most widely spoken of the Eastern Romance branch), which presents a consistent degree of similarity to Aromanian, Megleno-Romanian, and Istro-Romanian, but shares many features equally with the rest of the Western Romance languages, specifically Italian, French, Spanish, Portuguese, and Catalan. The Romanian alphabet contains the same 26 letters of the standard Latin alphabet, as well as five additional ones (namely ă, â, î, ț, and ș), totalling 31. Romanian is spoken as a first language by 91.55% of the entire population, while Hungarian and Vlax Romani are spoken by 6.28% and 1.20% of the population, respectively. There are also 40,861 native speakers of Ukrainian (concentrated in some compact regions near the border, where they form local majorities), 17,101 native speakers of Turkish, 15,943 native speakers of German, and 14,414 native speakers of Russian living in Romania. According to the Constitution, local councils ensure linguistic rights to all minorities. In localities with ethnic minorities of over 20%, that minority's language can be used in the public administration, justice system, and education. Foreign citizens and stateless persons who live in Romania have access to justice and education in their own language. English and French are the main foreign languages taught in schools. In 2010, the Organisation internationale de la Francophonie identified 4,756,100 French speakers in the country.
In Sri Lanka, there are seventeen public universities. Most public universities are funded by the government through the University Grants Commission, which handles undergraduate placements and staff appointments. The top institutions include the University of Peradeniya founded in 1942 and the University of Colombo founded in 1921. Sri Lanka also has a joint service military university, the General Sir John Kotelawala Defence University, which is operated by the Ministry of Defence.
Glutathione S-transferase A2 is an enzyme that in humans is encoded by the GSTA2 gene. Cytosolic and membrane-bound forms of glutathione S-transferase are encoded by two distinct supergene families. These enzymes function in the detoxification of electrophilic compounds, including carcinogens, therapeutic drugs, environmental toxins and products of oxidative stress, by conjugation with glutathione. The genes encoding these enzymes are known to be highly polymorphic. These genetic variations can change an individual's susceptibility to carcinogens and toxins as well as affect the toxicity and efficacy of some drugs. At present, eight distinct classes of the soluble cytoplasmic mammalian glutathione S-transferases have been identified: alpha, kappa, mu, omega, pi, sigma, theta and zeta. This gene encodes a glutathione S-transferase belonging to the alpha class. The alpha class genes, located in a cluster mapped to chromosome 6, are the most abundantly expressed glutathione S-transferases in liver. In addition to metabolizing bilirubin and certain anti-cancer drugs in the liver, the alpha class of these enzymes exhibit glutathione peroxidase activity thereby protecting the cells from reactive oxygen species and the products of peroxidation.
ATC code D Dermatologicals is a section of the Anatomical Therapeutic Chemical Classification System, a system of alphanumeric codes developed by the World Health Organization (WHO) for the classification of drugs and other medical products. Codes for veterinary use (ATCvet codes) can be created by placing the letter Q in front of the human ATC code: for example, QD. ATCvet codes without corresponding human ATC codes are cited with the leading Q in the following list.National versions of the ATC classification may include additional codes not present in this list, which follows the WHO version.
Chick-fil-A was first introduced into the Canadian market in 1981. They opened their first locations in Oakville Place, and Burlington Centre in Ontario. These locations would later close, however it is not clear when they were shuttered. In September 1994, Chick-fil-A re-entered Canada inside a student centre food court at the University of Alberta in Edmonton, Alberta. This location did not perform very well and was closed within two or three years. The company opened an outlet at the Calgary International Airport in Alberta in May 2014, and closed it in 2019. Chick-fil-A opened a restaurant in Toronto, Ontario, on September 6, 2019, in the Yonge and Bloor Street area. There were protests criticizing the company's violation of animal rights and "history of supporting anti-LGBTQ causes". Chick-fil-A announced that it would open two other locations in Toronto during 2019, and 12 additional stores in the Greater Toronto Area over the subsequent five years. The chain's second Toronto location opened at the Yorkdale Shopping Centre in January 2020. The company expanded to other areas of Ontario in 2021, opening standalone locations with drive-through restaurants in Kitchener in August and Windsor in October. In November 2023, the first standalone with drive-through location in London was opened on Wonderland Road South. In 2024, the company opened locations in Alberta, one being in the Phase III food court of West Edmonton Mall in Edmonton. and the other being in Calgary located on Macleod Trail.

After 1945, many of the films of the immediate post-war period can be characterised as Trümmerfilm (rubble film). East German film was dominated by the state-owned film studio DEFA, while the dominant genre in West Germany was the Heimatfilm ("homeland film"). The Academy Award for Best Foreign Language Film ("Oscar") went to the German production The Tin Drum (Die Blechtrommel) in 1979, to Nowhere in Africa (Nirgendwo in Afrika) in 2002, and to The Lives of Others (Das Leben der Anderen) in 2007. Various Germans won an Oscar for their performances in films. The annual European Film Awards ceremony is held every other year in Berlin, home of the European Film Academy. The Berlin International Film Festival known as "Berlinale", awarding the "Golden Bear" and held annually since 1951, is one of the world's leading film festivals. The "Lolas" are annually awarded in Berlin, at the German Film Awards.
== Regenerative medicine applications == The MRL/MpJ mouse has attracted considerable attention in the field of regenerative medicine owing to its remarkable capacity for tissue repair in the absence of fibrosis. Insights derived from this model have been investigated for their potential translational relevance to human therapies. One major area of interest is scarless wound healing. The ability of MRL/MpJ mice to regenerate injured tissues without forming scar tissue provides a valuable model for elucidating mechanisms that may help reduce fibrosis in human cutaneous injuries and post-surgical healing. Cardiac regeneration represents another promising avenue. Experimental studies have demonstrated that MRL/MpJ mice are capable of partial myocardial regeneration following injury, suggesting potential strategies for promoting cardiac repair after myocardial infarction. Furthermore, this strain has been employed to investigate mechanisms underlying skeletal muscle repair. Its relative resistance to muscular degeneration has provided insights into potential therapeutic approaches for neuromuscular disorders, including muscular dystrophy. In addition, studies of the MRL/MpJ model have contributed to understanding the role of metabolic regulation in tissue repair. Notably, resistance to diet-induced hyperglycemia in this strain may offer insights into improving wound healing in diabetic conditions.
Willard Marriott gained franchise rights for Washington, D.C., Baltimore and Richmond. Subsequently, he moved to Washington to open a root beer stand with Hugh Colton. These stands subsequently became Hot Shoppes, paving the way for the later creation of one of the largest hospitality corporations in the world. As of 1933, A&W had a total of 171 stands, increasing to 260 by 1941. A&W survived the Great Depression and labor shortages during World War II but benefitted from the post-war recovery of the American economy, with fast food and drive-in restaurants beginning to take shape. The first franchise convention was held in La Crosse, Wisconsin in 1949, but was limited to Wisconsin franchisees.
Although a majority of Millennials and Generation Z would like to get married one day, two fifths deem marriage to be an outdated institution and an overwhelming majority think it is unnecessary for a fulfilling or happy life. Moreover, they generally prefer an equitable partnership to a traditional patriarchal arrangement. Surveys have also documented a marked decline in the desire for children. Close to a quarter of Millennials and Generation Z say they do not want children. Many of them are seeking to be sterilization procedures (vasectomies for men and tubal ligation for women), an uptick most pronounced in politically conservative states, where abortion, and even contraception and sterilization, could be restricted or banned following the Supreme Court decision Dobbs v. Jackson (2022). Some have chosen to have pets in lieu of children, and they frequently refer to these animals as members of their families or their own children ("fur babies"). Generation Z prefer to prioritize professional life and individual self-discovery, are concerned with financial constraints driven by high inflation and taxes, and dislike the perceived constraints of family life. Those who wish to be parents want to achieve financial security and to be in a serious relationship first.

In ambient ionization, ions are formed outside the mass spectrometer without sample preparation or separation. Ions can be formed by extraction into charged electrospray droplets, thermally desorbed and ionized by chemical ionization, or laser desorbed or ablated and post-ionized before they enter the mass spectrometer. Solid-liquid extraction based ambient ionization uses a charged spray to create a liquid film on the sample surface. Molecules on the surface are extracted into the solvent. The action of the primary droplets hitting the surface produces secondary droplets that are the source of ions for the mass spectrometer. Desorption electrospray ionization (DESI) creates charged droplets that are directed at a solid sample a few millimeters to a few centimeters away. The charged droplets pick up the sample through interaction with the surface and then form highly charged ions that can be sampled into a mass spectrometer. Plasma-based ambient ionization is based on an electrical discharge in a flowing gas that produces metastable atoms and molecules and reactive ions. Heat is often used to assist in the desorption of volatile species from the sample. Ions are formed by chemical ionization in the gas phase. A direct analysis in real time (DART) source operates by exposing the sample to a dry gas stream (typically helium or nitrogen) that contains long-lived electronically or vibronically excited neutral atoms or molecules (or "metastables"). Excited states are typically formed in the DART source by creating a glow discharge in a chamber through which the gas flows.

Viserys had Jace and Daeron share the same wetnurse to bring Alicent and Rhaenyra closer, while denying the accusations that Rhaenyra's sons were bastards fathered by Ser Harwin Strong instead of Laenor. Daemon and his new wife Laena had twin daughters Baela and Rhaena Targaryen in 116 AC, as Viserys reunited with his brother once again. To improve their relations, Viserys gave his blessing to betrothe Jace to Baela and Luke to Rhaena. Several tragedies occurred in 120 AC, beginning with Laena's death following a failed birth. Shorty after, Laenor was killed by his rumored lover Ser Qarl Correy in a duel at Driftmark. At his funeral, Aemond got into a violent fight with Jace, Luke and Joffrey after he called the brothers "bastards", which culminated in Aemond losing an eye when Luke attacked him with a knife. This inflamed tensions between the greens and blacks, whose apologies to one another were taken seriously by no one but Viserys. He ordered Harwin to depart Dragonstone for Harrenhal, and issued an edict that anyone who spread rumors of Rhaenyra's sons' illegitimacy would have their tongue cut out. Harwin and his father Lyonel Strong would soon perish in a fire at Harrenhal, with Viserys considered one of the suspects of setting the fire. To replace Lyonel, Viserys recalled Otto to serve him a second time as his Hand. As both Daemon and Rhaenyra were now widows, the two married each other six months later without Viserys' knowledge, which was a large scandal in court.

Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
Albumin is the most abundant protein in plasma and carries molecules that bear fatty-acid chains. Binding shields the peptide from renal filtration and from peptidases, keeping a circulating reservoir. Slow release from this reservoir produces sustained receptor occupancy and supports infrequent dosing.
The insulinotropic effect is glucose-dependent, meaning secretion increases mainly when glucose is elevated. This property is often described as lowering the chance of hypoglycaemia when the compound is used alone. Other glucose-lowering agents used at the same time can still cause low blood glucose.
It is a synthetic analog of GLP-1 produced through medicinal chemistry to resist enzymatic degradation. The design goal was longer circulation than the native hormone.
