Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.

The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
Thyroid hormones are two hormones produced and released by the thyroid gland: triiodothyronine (T3) and thyroxine (T4). They are tyrosine-based hormones that are primarily responsible for regulation of metabolism. T3 and T4 are partially composed of iodine, which is derived from food. A deficiency of iodine leads to decreased production of T3 and T4, enlarges the thyroid tissue, and causes the disease known as simple goitre. The major form of thyroid hormone in the blood is thyroxine (T4), whose half-life of around one week is longer than that of T3. In humans, the ratio of T4 to T3 released into the blood is approximately 14:1. T4 is converted to the active T3 (three to four times more potent than T4) within cells by deiodinases (5′-deiodinase). These are further processed by decarboxylation and deiodination to produce iodothyronamine (T1a) and thyronamine (T0a). All three isoforms of the deiodinases are selenium-containing enzymes, thus dietary selenium is essential for T3 production. Calcitonin, a peptide hormone produced and secreted by the thyroid, is usually not included in the meaning of "thyroid hormone". Thyroid hormones are one of the factors responsible for the modulation of energy expenditure. This is achieved through several mechanisms, such as mitochondrial biogenesis and adaptive thermogenesis. American chemist Edward Calvin Kendall was responsible for the isolation of thyroxine in 1915. In 2020, levothyroxine, a manufactured form of thyroxine, was the second most commonly prescribed medication in the United States, with more than 98 million prescriptions.
==== Protease inhibitors ==== Some viruses include an enzyme known as a protease that cuts viral protein chains apart so they can be assembled into their final configuration. HIV includes a protease, and so considerable research has been performed to find "protease inhibitors" to attack HIV at that phase of its life cycle. Protease inhibitors became available in the 1990s and have proven effective, though they can have unusual side effects, for example causing fat to build up in unusual places. Improved protease inhibitors are now in development. Protease inhibitors have also been seen in nature. A protease inhibitor was isolated from the shiitake mushroom (Lentinus edodes). The presence of this may explain the Shiitake mushrooms' noted antiviral activity in vitro.
== Principle == This technique is applicable to gases or any materials that can be vaporized. It uses a non-fragmenting non-conventional ionisation mode, by attachment of a lithium (or alkaline) ion to the gas to be analysed with a more traditional mass filter. This instrument is more dedicated to analysis of moderately-sized molecules such as organic or aromatic compounds.

In 1964, the first franchise was purchased by former police officer Kermit Bekke. This location, located at 1654 West Carson Street in Torrance, California, had its grand opening on May 28, 1965. Bekke sold his franchise a year and a half later. This location closed in 1975 when it moved across the street to store #1130 at 1619 West Carson Street, which itself would close in the late 1990s and subsequently be repurposed. On January 20, 1967, the 100th Taco Bell grand opening took place in Anaheim, located at 400 South Brookhurst (store #63); this location would later be replaced by a new location at 324 South Brookhurst in 1993. The first Taco Bell restaurant east of the Mississippi River (the 270th to open, store #258) opened at 2050 East Main Street in Springfield, Ohio, on September 20, 1968. Original Taco Bells featured only walk-up window service without indoor seating or drive-thru service. Inside seating was added sporadically in 1968 and drive-thru service was not generally available until 1980. As of 2025, seven original mission-style operating Taco Bell restaurants still remain, located in California and Colorado. A mission-style location (opened in April 1976) that operated in Honolulu, Hawaii (store #1281) remodeled in 2024, and another Mission-style location in Scottsdale, Arizona (store #31, opened circa July 3, 1966) closed on April 12, 2025.
== Pharmacology == Sunobinop has nanomolar affinity (Ki) and efficacy (EC50) at human recombinant nociceptin/orphanin-FQ peptide (NOP) receptors. It has a high degree of functional selectivity for the NOP receptor. Sunobinop is a low affinity antagonist at human mu and kappa opioid receptors, and is a low affinity weak partial agonist at human delta opioid receptors.
== Opposition to atomic theory == Dalton's atomic theory attracted widespread interest but not universal acceptance. One problem was the lack of uniform nomenclature. The word "atom" implied indivisibility, but Dalton defined an atom as being the ultimate particle of any chemical substance, not just the elements or even matter per se. This meant that "compound atoms" such as carbon dioxide could be divided, as opposed to "elementary atoms". Dalton disliked the word "molecule", regarding it as "diminutive". Amedeo Avogadro did the opposite: he exclusively used the word "molecule" in his writings, eschewing the word "atom", instead using the term "elementary molecule". Jöns Jacob Berzelius used the term "organic atoms" to refer to particles containing three or more elements, because he thought this only existed in organic compounds. Jean-Baptiste Dumas used the terms "physical atoms" and "chemical atoms"; a "physical atom" was a particle that cannot be divided by physical means such as temperature and pressure, and a "chemical atom" was a particle that could not be divided by chemical reactions. The modern definitions of atom and molecule—an atom being the basic particle of an element, and a molecule being an agglomeration of atoms—were established in the latter half of the 19th century. A key event was the Karlsruhe Congress in Germany in 1860. As the first international congress of chemists, its goal was to establish some standards in the community. A major proponent of the modern distinction between atoms and molecules was Stanislao Cannizzaro.

=== Charges === On January 7th, 2010, U.S. Immigration and Customs Enforcement (ICE) agents authorized by a grand jury with a search and arrest warrant entered the residence of Mahmoud Reza Banki in New York City and arrested him. He was arraigned before Judge John F. Keenan on an indictment charging him with three counts: 1. Conspiracy to violate the U.S.-imposed Iran sanctions and conspiracy to run an unlicensed money transmittal system, 2. Violation of the Iran sanctions, and 3. Running of an unlicensed money transmittal business. The indictment accused Banki of receiving $4.7 million in violation of the Iran sanctions. Prosecutors filed a superseding indictment later changing the amount to $3.4 million and adding two false statement charges to the three initial charges.
=== Provincial government === Yasir Naqvi, the Attorney General of Ontario, and Eric Hoskins, the province's Minister of Health and Long-Term Care, jointly announced on the day of Wettlaufer's sentencing that the provincial government would commission a public inquiry into her case. The full details of the inquiry were not given in the announcement, as the government had yet to determine the scope or an individual to lead the inquiry, with Naqvi and Hoskins instead saying that the inquiry would "get the answers we need to help ensure a tragedy such as this does not happen again." The delay in establishing the inquiry was criticized by members of the opposition Progressive Conservative and New Democratic parties toward the end of July 2017, as no progress had seemingly been made since the announcement and the Legislative Assembly had risen for its summer recess. The Public Inquiry into the Safety and Security of Residents in the Long-Term Care Homes System was formally established by the provincial government on August 1, 2017. Justice Eileen Gillese of the Court of Appeal for Ontario was appointed commissioner of the inquiry. The inquiry was to include interviews with victims' families and public consultations in the community as it investigated the circumstances surrounding the deaths of Wettlaufer's victims and gaps in legislative or policy frameworks that allowed her to continue working as a nurse. The inquiry's lead counsel stated that "anyone from Wettlaufer to Premier Kathleen Wynne" may be called to testify before the inquiry based on the evidence that is uncovered.
Nicotinamide N-methyltransferase (NNMT) is an enzyme that in humans is encoded by the NNMT gene. NNMT catalyzes the methylation of nicotinamide and similar compounds using the methyl donor S-adenosyl methionine (SAM-e) to produce S-adenosyl-L-homocysteine (SAH) and 1-methylnicotinamide.
== Sunderland's classification == In 1951, Sunderland expanded Seddon's classification to five degrees. The first two are the same as Seddon's. With each degree, the severity of the injury increases and has larger structural disruption and poorer prognosis. Sunderland's third-degree and fourth-degree are included within Seddon's axonotmensis. Sunderland's third-degree is nerve fiber interruption. This includes an endoneurium lesion with disruption of the axon and endoneurium, but an intact epineurium and perineurium. Recovery from a third-degree injury may require surgical intervention due to misdirected regeneration that is caused by endoneurial damage. In fourth-degree injury, only the epineurium remain intact, and a complete block to axonal regeneration could occur, requiring surgical repair. Sunderland's fifth-degree is included within Seddon's neurotmesis. Fifth-degree lesion is a complete transection of the nerve, including the epineurium. Recovery requires appropriate surgical treatment since there is no spontaneous recovery expected.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
