=== Candidacy for mayorship of Porto Alegre in 2016 === In 2016, Melo was the candidate for the mayorship of PMDB, with state deputy Juliana Brizola, of the PDT, as the vice-mayoral candidate on the ticket. In the first round, they received 185,655 votes (25.93%). They came in second place behind Nelson Marchezan Júnior, and both qualified for the second round. Melo received a diverse array of support, including from the PCdoB, the PMN, and from the PV's candidate, Marcelo Chiodo, even as his party supported Marchezan Júnior. He also positioned himself as more to the left, announcing his support for the fight against PEC 241, which established a ceiling for public spending for twenty years in Brazil, also going against the national position of his party. Along with this, he affirmed his position against proposed laws inspired by Escola sem Partido, a conservative movement. Melo declared that he had an "identity of struggles" with the left, since he came from the "old MDB", as the party was the main political force against the military dictatorship. As a candidate, he had shown opposition against the privatization of public companies, such as DMAE, Procempa and Carris, and defended participatory budgeting. After a tense campaign with constant trading of accusations between him and Marchezan Júnior, Melo was defeated in the second round, receiving 262,601 votos (39.50%). After the election, on analyzing his defeat in an interview, Melo criticized the possibility that the MDB would become a part of his adversary's administration.
=== Comparison with other expression systems === In standard molecular biology research, the bacterium Escherichia coli is the most frequently used organism for expression system, to produce heterologous proteins, due to its features of fast growth rate, high protein production rate, as well as undemanding growth conditions. Protein production in E. coli is usually faster than that in Komagataella, with reasons: Competent E. coli cells can be stored frozen, and thawed before use, whereas Komagataella cells have to be produced immediately before use. Expression yields in Komagataella vary between different clones, so that a large number of clones has to be screened for protein production, to find the best producer. The biggest advantage of Komagataella over E. coli is that Komagataella is capable of forming disulfide bonds and glycosylations in proteins, but E. coli cannot. E. coli might produce a misfolded protein when disulfides are included in final product, leading to inactive or insoluble forms of proteins. The well-studied Saccharomyces cerevisiae is also used as an expression system with similar advantages over E. coli as Komagataella. In 2023, the United States saw a record in crude oil production with over 13.2 million barrels of crude per day, beating the 13 million barrels per day produced at the peak of Trump's presidency. The United States also dealt with supply shocks caused by the 2021–2024 global energy crisis due to the COVID-19 pandemic and Russian invasion of Ukraine. An advocate for environmental justice to address the impact of climate change on lower-income areas and people of color, Harris supported Biden's climate legislation. In 2022, Harris helped pass the Inflation Reduction Act, the largest investment in addressing climate change and clean energy in American history, putting the United States on track to meet emissions reduction targets by 50–52% below 2005 levels by 2030. Harris's campaign stated that she would not support a ban on fracking. Trump ridiculed the idea of man-made climate change, and repeatedly referred to his energy policy under the mantra "drill, baby, drill". Huntington's disease is a trinucleotide repeat disorder caused by trinucleotide repeat expansion in the first exon of the huntingtin gene (HTT), which encodes the huntingtin protein (HTT). HTT may also be referred to as the HD gene or, historically, IT15 (interesting transcript 15). It is located on the short arm of chromosome 4 at 4p16.3. The trinucleotide repeat region of HTT is primarily composed of CAG, which encodes the amino acid glutamine; thus, the resulting protein contains a polyglutamine tract (polyQ tract). The number of repeats varies in length between individuals and may change length between generations. When the length of this repeated section exceeds a certain threshold, it produces mutant huntingtin protein (mHTT). In turn, mHTT has toxic gains and losses of function which negatively impact cell function and lead to disease. The Huntington's disease mutation is genetically dominant and almost fully penetrant; a single mutant HTT allele from either parent is sufficient to cause the disease. Because the rate of mutation is higher in sperm cells, males are more likely to transmit expanded HTT alleles to their offspring.

== Causes == A spontaneous spinal CSF leak refers to any CSF leak that has not been preceded by a surgical or other medical procedure. Later evidence suggests that these leaks result from either a discogenic pathology, such as microspur, osteophyte or spinal disc herniation that pierces the dura like a knife, connective tissue disorder (which can often lead to discogenic pathology), or spinal drainage problems. A cerebrospinal fluid leak can be a rare complication of an anterior cervical discectomy and fusion. One study suggested a CSF leak to follow from 0.5% of operations. Another study suggests a CSF leak to follow from 1% of operations. In most of these cases repair is successful.

Tests such as chest X-rays can be used to explore and exclude alternate causes of a person's symptoms. Echocardiography may assist in modifying clinical suspicion of ongoing myocardial infarction in patients that can't be ruled out or ruled in following initial ECG and Troponin testing. Myocardial perfusion imaging has no role in the acute diagnostic algorithm; however, it can confirm a clinical suspicion of Chronic Coronary Syndrome when the patient's history, physical examination (including cardiac examination) ECG, and cardiac biomarkers suggest coronary artery disease. Echocardiography, an ultrasound scan of the heart, is able to visualize the heart, its size, shape, and any abnormal motion of the heart walls as they beat that may indicate a myocardial infarction. The flow of blood can be imaged, and contrast dyes may be given to improve image. Other scans using radioactive contrast include SPECT CT-scans using thallium, sestamibi (MIBI scans) or tetrofosmin; or a PET scan using Fludeoxyglucose or rubidium-82. These nuclear medicine scans can visualize the perfusion of heart muscle. SPECT may also be used to determine viability of tissue, and whether areas of ischemia are inducible.

In February 2013, proteogenomic mapping research was done with ENCODE to identify translational regions in the human genome. They applied peptide fingerprint scanning and MASCOT to the protein data to find regions that may not have been previously annotated as translated in the human genome. This search against the whole genome revealed that approximately 4% of unique peptide that they found were outside of previously annotated regions. Also the comparison of the whole genome revealed 15% more hits than from a protein database search (such as MASCOT) alone. GFS can be used as a complementary method for annotation due to the fact that you can find new genes or splice sites that have not been annotated before. However it is important to remember that the whole genome approach used by GFS can be less sensitive than programs that look only at annotated regions.
