GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L cells after food intake. The molecule is a 31-amino-acid backbone modified at three positions to resist cleavage by dipeptidyl peptidase-4, the enzyme that degrades native GLP-1 within minutes. A lysine residue at position 26 carries a linker and a C18 fatty diacid, which promotes binding to serum albumin and slows renal clearance. These changes extend the circulating half-life from roughly two minutes to about one week in humans.
The sequence incorporates alpha-aminoisobutyric acid at position 8, replacing the alanine found in the natural hormone. This substitution blocks the primary DPP-4 recognition site and contributes most of the enzymatic stability. Albumin binding further protects the peptide and reduces the frequency of administration required to maintain active plasma levels. Because the fatty acid chain increases lipophilicity, the compound is formulated as a solution rather than a simple aqueous buffer. Researchers describe the design as an incremental optimization of earlier GLP-1 analogs rather than a wholly new scaffold.

Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Five Guys Enterprises, LLC (doing business as Five Guys Burgers and Fries and Five Guys) is an American multinational fast food chain focused on hamburgers, hot dogs, and french fries. It is headquartered in Alexandria, Virginia. The first Five Guys restaurant opened in 1986 in Arlington County, Virginia. By 2001, there were five locations in the Washington, D.C., metro area. In early 2003, Five Guys began franchising, beginning a period of rapid expansion. In a year and a half, permits had been sold for over 300 franchised locations. As of 2026, Five Guys had over 1,900 locations open worldwide. It was the fastest-growing fast food chain in the United States, with a 32.8% sales increase from 2010 to 2011.
Latex can trigger an IgE-mediated cutaneous, respiratory, and systemic reaction. The prevalence of latex allergy in the general population is believed to be less than one percent. In a hospital study, 1 in 800 surgical patients (0.125 percent) reported latex sensitivity, although the sensitivity among healthcare workers is higher, between seven and ten percent. Researchers attribute this higher level to the exposure of healthcare workers to areas with significant airborne latex allergens, such as operating rooms, intensive-care units, and dental suites. These latex-rich environments may sensitize healthcare workers who regularly inhale allergenic proteins. The most prevalent response to latex is allergic contact dermatitis, a delayed hypersensitivity reaction that manifests as dry, crusted lesions. This reaction usually lasts 48–96 hours. Sweating or rubbing the area under the glove aggravates the lesions, possibly leading to ulcerations. Anaphylactic reactions occur most often in sensitive patients who have been exposed to a surgeon's latex gloves during abdominal surgery, but other mucosal exposures, such as dental procedures, can also produce systemic reactions. Latex and banana sensitivity may cross-react. Furthermore, those with latex allergy may also have sensitivities to avocado, kiwifruit, and chestnut. These people often have perioral itching and local urticaria. Only occasionally have these food-induced allergies induced systemic responses.
35Cl + n → 35S + p It decays by beta-decay with a half-life of 87.51 days. It is used to label the sulfur-containing amino-acids methionine and cysteine. When a sulfur atom replaces an oxygen atom in a phosphate group on a nucleotide a thiophosphate is produced, so 35S can also be used to trace a phosphate group.
== Membrane separation processes == Membrane separation processes have a very important role in the separation industry. Nevertheless, they were not considered technically important until the mid-1970s. Membrane separation processes differ based on separation mechanisms and size of the separated particles. The widely used membrane processes include microfiltration, ultrafiltration, nanofiltration, reverse osmosis, electrolysis, dialysis, electrodialysis, gas separation, vapor permeation, pervaporation, membrane distillation, and membrane contactors. All processes except for pervaporation involve no phase change. All processes except electrodialysis are pressure driven. Microfiltration and ultrafiltration is widely used in food and beverage processing (beer microfiltration, apple juice ultrafiltration), biotechnological applications and pharmaceutical industry (antibiotic production, protein purification), water purification and wastewater treatment, the microelectronics industry, and others. Nanofiltration and reverse osmosis membranes are mainly used for water purification purposes. Dense membranes are utilized for gas separations (removal of CO2 from natural gas, separating N2 from air, organic vapor removal from air or a nitrogen stream) and sometimes in membrane distillation. The later process helps in the separation of azeotropic compositions reducing the costs of distillation processes.
=== PAM === One of the first amino acid substitution matrices, the PAM (Point Accepted Mutation) matrix was developed by Margaret Dayhoff in the 1970s. This matrix is calculated by observing the differences in closely related proteins. Because the use of very closely related homologs, the observed mutations are not expected to significantly change the common functions of the proteins. Thus the observed substitutions (by point mutations) are considered to be accepted by natural selection. One PAM unit is defined as 1% of the amino acid positions that have been changed. To create a PAM1 substitution matrix, a group of very closely related sequences with mutation frequencies corresponding to one PAM unit is chosen. Based on collected mutational data from this group of sequences, a substitution matrix can be derived. This PAM1 matrix estimates what rate of substitution would be expected if 1% of the amino acids had changed. The PAM1 matrix is used as the basis for calculating other matrices by assuming that repeated mutations would follow the same pattern as those in the PAM1 matrix, and multiple substitutions can occur at the same site. With this assumption, the PAM2 matrix can estimated by squaring the probabilities. Using this logic, Dayhoff derived matrices as high as PAM250. Usually the PAM 30 and the PAM70 are used.

Later during the war, Kitchener attempted to form a Boer Police Force, as part of his efforts to pacify the occupied areas and effect a reconciliation with the Boer community. The members of this force were despised as traitors by the Boers still in the field. Boers who attempted to remain neutral after giving their parole to British forces were derided as "hensoppers" (hands-uppers) and often coerced into giving support to the Boer guerrillas (which was one reason for British scorched earth campaigns throughout the countryside and detention of Boers in concentration camps, to deny anything of use to the guerrillas). Like the Canadian, and particularly the Australian and New Zealand contingents, many volunteer units formed by South Africans were "light horse" or mounted infantry, well-suited to the countryside and manner of warfare. Some regular British officers scorned their comparative lack of formal discipline, but the light horse units were hardier and more suited to campaigning than the overloaded British cavalry, who were still obsessed with the charge by lance or sabre. At their peak, 24,000 South Africans served in the field in "colonial" units. Notable units (in addition to the Imperial Light Horse) were the South African Light Horse, Rimington's Guides, Kitchener's Horse and the Imperial Light Infantry.
== Low-level exposure == As the energy of radio frequency waves and microwaves is insufficient to directly disrupt individual chemical bonds in small or stable molecules, the effects are considered limited to thermal. Energy densities that are not sufficient to overheat the tissues are not shown to cause lasting damage. To clarify, the deep-red lightbulb in a black-and-white photographic darkroom produces a higher-energy form of radiation than microwaves. Like a microwave, this lightbulb can burn, particularly if touched, but the burn is only possible due to too much heat. A study of 20,000 radar technicians of the US Navy, who were chronically exposed to high levels of microwave radiation, did not detect increased incidence of cancer. Recent epidemiologic evidence also led to the consensus that exposure to electromagnetic fields, e.g. along power lines, did not raise incidence of leukemia or other cancers.
==== Dol ==== The dol (from the Latin word for pain, dolor) is a unit of measurement for pain. James D. Hardy, Herbert G. Wolff, and Helen Goodell of Cornell University proposed the unit based on their studies of pain during the 1940s and 1950s. They defined one dol to equal a just-noticeable difference in pain. The unit never came into widespread use and other methods are now used to assess the level of pain experienced by patients.
==== Extreme hunger ==== After experiencing prolonged significant calorie deficits, people often undergo extreme hunger (hyperphagia). Once weight is regained in both fat and muscle mass, their hunger usually returns to normalized levels. This process of regaining weight takes longer for fat-free/muscle mass which leads to a "fat overshoot" or "overshoot weight". This causes patient's body fat levels to be greater than before the disorder. This kind of process for hunger normalization varies between patient and could last anywhere from a few months to multiple years.

=== Location === Tarlov cysts are most commonly located in the S1 to S4/S5 region of the spinal canal, but can be found along any region of the spine. Tarlov cysts typically form on the extradural portions of the sacral nerve roots, most often near the dorsal root ganglion of the dorsal (sensory) nerve roots. They arise within the perineural space, between the endoneurium and perineurium. Occasionally, these cysts are observed in the lumbar and thoracic spine. The cysts are often multiple, extending around the circumference of the nerve, and can enlarge over time to compress neighboring nerve roots, to cause bone erosion. The cysts may also form on the anterior (front) side of the sacrum and have been known to extend into the pelvic cavity. These cysts, though rare, can be found to grow large - over 3–4 centimetres (1.2–1.6 in) in size, often causing severe pelvic pain from compression on the cyst itself as well as adjoining nerves.
=== Maximum-likelihood methods === The maximum-likelihood approach uses probability theory to complete all three steps simultaneously. It estimates critical parameters, including the divergence between sequences and the transition/transversion ratio, by deducing the most likely values to produce the input data.
All of astatine's isotopes are short-lived; the most stable is astatine-210, with a half-life of 8.1 hours. There are 41 known isotopes of astatine, with mass numbers of 188 and 190–229. Theoretical modeling suggests that about 37 more isotopes could exist. No stable or long-lived astatine isotope has been observed, nor is one expected to exist. Astatine's alpha decay energies follow the same trend as for other heavy elements. Lighter astatine isotopes have quite high energies of alpha decay, which become lower as the nuclei become heavier. Astatine-211 has a significantly higher energy than the previous isotope, because it has a nucleus with 126 neutrons, and 126 is a magic number corresponding to a filled neutron shell. Despite having a similar half-life to the previous isotope (8.1 hours for astatine-210 and 7.2 hours for astatine-211), the alpha decay probability is much higher for the latter: 41.81% against only 0.18%. The two following isotopes release even more energy, with astatine-213 releasing the most energy. For this reason, it is the shortest-lived astatine isotope. Even though heavier astatine isotopes release less energy, no long-lived astatine isotope exists, because of the increasing role of beta decay (electron emission). This decay mode is especially important for astatine; as early as 1950 it was postulated that all isotopes of the element undergo beta decay, though nuclear mass measurements indicate that 215At is in fact beta-stable, as it has the lowest mass of all isobars with A = 215.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
