The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.

Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
The output of the device was 1.018 volts and was held to within a few parts per million. The principle of the vacuum flask makes it ideal for storing certain types of rocket fuel, and NASA used it extensively in the propellant tanks of the Saturn launch vehicles in the 1960s and 1970s. The design and shape of the Dewar flask was used as a model for optical experiments based on the idea that the shape of the two compartments with the space in between is similar to the way the light hits the eye. The vacuum flask has also been part of experiments using it as the capacitor of different chemicals in order to keep them at a consistent temperature. The industrial Dewar flask is the base for a device used to passively insulate medical shipments. Most vaccines are sensitive to heat and require a cold chain system to keep them at stable, near freezing temperatures. The Arktek device uses eight one-litre ice blocks to hold vaccines at under 10 °C. In the oil and gas industry, Dewar flasks are used to insulate the electronic components in wireline logging tools. Conventional logging tools (rated to 350 °F) are upgraded to high-temperature specifications by installing all sensitive electronic components in a Dewar flask.
In the mid-afternoon on September 2, in the Ana Kalay Valley, following a similar ruse that killed 7 insurgents, 39 Green Berets, SASR soldiers and Afghan Police on five GMVs were returning to an American patrol base when they were engaged by small arms and RPG fire from four firing points, starting the Battle of Khaz Oruzgan. One Green Beret was seriously wounded, a U.S. JTAC called in a flight of F/A-18 Hornets that conducted gun runs against the insurgents and returned to drop JDAMs on a group of insurgents, as the patrol moved with their vehicles more troops were hit, they eventually arrived at the patrol base, of the 13 wounded- 7 were SASR soldiers and a Green Beret was killed; one SASR soldier was awarded the Victoria Cross for his actions during the battle.
The white shark is difficult to keep in captivity due to its large size and migratory nature. Attempts have been made since 1955 in facilities in North America, Hawaii, Australia, and South Africa. The sharks survived only for days during the earliest attempts, while in the early 1980s, aquariums like Steinhart Aquarium, Sea World San Diego, and Marineland of the Pacific were able to keep juvenile white sharks for weeks before releasing them. A major contributor to the mortality of captive white sharks was the stress of poor transport; many were accidentally captured by commercial nets and kept on fishing lines or in a tank before being handed over to aquarium staff. One famous shark named Sandy, who measured 2.3 m (7.5 ft), was kept at Steinhart for five days in August 1980 and was released after bumping into the walls. The most successful exhibitions occurred at Monterey Bay Aquarium (MBA), which displayed six white sharks between 2004 and 2011. Researchers at universities in California attributed this success to a 4-million-US-gallon (15,000,000 L) net pen, which gave the sharks time to recover from capture before being moved. Additionally, a 3,200-US-gallon (12,000 L) portable tank allowed the sharks to swim continuously during transit. Although the sharks were 1.4–1.6 m (4.6–5.2 ft) at the start of their stay, they eventually outgrew the exhibit and required release. One shark was kept for 198 days and attracted one million visitors. Having gathered sufficient data on the species, MBA discontinued the program in 2011.

In 2013, Chinese authorities announced that, since 2007, GSK had funnelled HK$3.8 billion in kickbacks to GSK managers, doctors, hospitals and others who prescribed their drugs, using over 700 travel agencies and consulting firms. Chinese authorities arrested four GSK executives as part of a four-month investigation into claims that doctors were bribed with cash and sexual favours. In 2014, a Chinese court found the company guilty of bribery and imposed a fine of US$490 million. Mark Reilly, the British head of GSK's Chinese operations, received a three-year suspended prison sentence after a one-day trial held in secret. Reilly was reportedly deported from China and dismissed by the company.
=== Earlier stages === Rifamycin inhibits bacterial DNA transcription into mRNA by inhibiting DNA-dependent RNA polymerase by binding its beta-subunit. alpha-Amanitin is a powerful inhibitor of eukaryotic DNA transcription machinery.
=== Nontoxicity for animals and humans === In different animal models, tuftsin showed no toxicity when administered intravenously or intraperitoneally. In a phase I study, tuftsin was shown to be nontoxic in adult human patients with advanced cancer when it was injected once intravenously (0.96 mg/kg body weight). Extensive augmentation of white blood counts and enhanced cytotoxicity of lymphocytes was notable. No detectable tuftsin-related toxicity was noticed in human patients during a phase II study, where the peptide was injected intravenously twice a week at total doses of 5 mg per injection.

"Controversy surrounds both the drug study itself and the FDA approval," wrote NEJM editor-at-large Richard P. Wenzel, MD in 2002. The FDA approved the drug despite the advisory committee's split vote (10 to 10) due to concerns about the validity of the claimed efficacy and safety findings on the basis of a single trial. Eli Lilly spokeswoman Judy Kay Moore insisted that the company did not mastermind the ethics task force or steer the guideline-writing process. It was only a coincidence, Moore says, that the ethics task force and the Surviving Sepsis Campaign used the same P.R. firm, Belsito and Company.

Marxism–Leninism–Maoism–Prachanda Path (Nepali: मालेमावाद र प्रचण्डपथ Mālemāvād ra Prachaṇḍapath; sometimes shortened to Prachanda Path) is the ideological line of the Communist Party of Nepal (Maoist Centre), also known as the CPN (MC). It is considered a development of Marxism–Leninism–Maoism (MLM) and named after the leader of the CPN (MC), Pushpa Kamal Dahal, commonly known as Prachanda. Prachanda Path was proclaimed in 2001. The ideology was partially inspired by the example of the Communist Party of Peru – Shining Path, which refers to its ideological line as Marxism–Leninism–Maoism–Gonzalo Thought. Prachanda Path does not claim to make an ideological break with Marxism, Leninism or Maoism, but rather to be an extension of these ideologies based on the politics of Nepal. The doctrine came into existence after the party determined that the ideologies of Marxism, Leninism and Maoism could no longer be practiced completely as they had been in the past. The party adopted Prachanda Path as they felt it was a suitable ideology based on the reality of Nepalese politics. Militarily and in the context of the 1996–2006 civil war in Nepal, central to the ideology was the achievement of revolution through the control of rural areas and the encirclement of urban settlements. The CPN (MC) was seen to have abandoned Maoism in its decision to merge with the Communist Party of Nepal (Unified Marxist–Leninist) to form the Nepal Communist Party, but this party has since been disestablished and the CPN (MC) reconstituted.
James Henry Wiggin, as an editor and literary adviser. The issue of how much Wiggin contributed to Science and Health is controversial. A former Unitarian clergyman, he was the book's editor from the 16th edition in 1886 until the 50th in 1891—22 editions appeared between 1886 and 1888 alone—and according to his literary executor, speaking after Wiggin's death, said he had rewritten it. Robert Peel wrote that Wiggin had "toned up" Eddy's style, but had not affected her thinking. In a letter to Wiggin in July 1886, Eddy wrote: "Never change my meaning, only bring it out." Eddy continued to revise the book until her death in 1910. In 1902 she added a chapter, "Fruitage," recounting healing testimonies from the Christian Science Journal and Christian Science Sentinel. There were over 400 editions (the final ran to 18 chapters and 600 pages), seven of them major revisions, according to Gottschalk, and members were encouraged to buy them all. Other income derived from the sale of rings and brooches, pictures of Eddy, and in 1889 the Mary Baker Eddy souvenir spoon; Eddy asked every Christian Scientist to buy at least one, or a dozen if they could afford to. When the copyright on Science and Health expired in 1971, the church persuaded Congress to extend it to 2046. The bill was supported by two of President Richard Nixon's aides, Christian Scientists H. R. Haldeman and John Ehrlichman. The law was overturned as unconstitutional in 1987, after a challenge by United Christian Scientists, an independent group. By 2001 Science and Health had sold over nine million copies.

It is a synthetic analog of GLP-1 produced through medicinal chemistry to resist enzymatic degradation. The design goal was longer circulation than the native hormone.
A fatty acid side chain attaches the peptide to serum albumin, which shields it from kidney filtration and protease activity. This interaction is the main reason its circulation time is extended.
No. The native hormone is GLP-1, and semaglutide is an engineered variant with three deliberate structural alterations. It does not appear in unmodified biological sources.
Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
