Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.

Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
== Lemna farming == High yields of Lemna with a high protein content for use in human nutrition, animal and fish feed can be achieved by careful control of growth conditions. Although Lemna can tolerate temperatures ranging from 6 °C (43 °F) to 33 °C (91 °F), the optimal growth range is 20 °C (68 °F) to 28 °C (82 °F)}. The acceptable pH range is 5 to 9, but better growth is obtained in the pH range of 6.5 to 7.5}. A minimum water depth of 1 foot (30 cm) is desirable to prevent excessive temperature swings. High nitrogen levels, for example 20 mM urea, have provided a protein content in the range of 45% by dry weight.
==== Preventing dehydration ==== Food and drink can help prevent dehydration in malnourished people with diarrhea. Eating (or breastfeeding, among infants) should resume as soon as possible. Sugary beverages like soft drinks, fruit juices, and sweetened teas are not recommended as they may worsen diarrhea. Malnourished people with diarrhea (especially children) should be encouraged to drink fluids; the best choices are fluids with modest amounts of sugar and salt, like vegetable broth or salted rice water. If clean water is available, they should be encouraged to drink that too. Malnourished people should be allowed to drink as much as they want, unless signs of swelling emerge. Babies can be given small amounts of fluids via an eyedropper or a syringe without the needle. Children under two should receive a teaspoon of fluid every one to two minutes; older children and adults should take frequent sips of fluids directly from a cup. After the first two hours, fluids and foods should be alternated, rehydration should be continued at the same rate or more slowly, depending on how much fluid the child wants and whether they are having ongoing diarrhea. If vomiting occurs, fluids can be paused for 5–10 minutes and then restarted more slowly. Vomiting rarely prevents rehydration, since fluids are still absorbed and vomiting is usually short-term.
One of the claimants of the English throne opposing William the Conqueror, Edgar Atheling, eventually fled to Scotland. King Malcolm III of Scotland married Edgar's sister Margaret, and came into opposition to William who had already disputed Scotland's southern borders. William invaded Scotland in 1072, riding as far as Abernethy where he met up with his fleet of ships. Malcolm submitted, paid homage to William and surrendered his son Duncan as a hostage, beginning a series of arguments as to whether the Scottish Crown owed allegiance to the King of England. Normans went into Scotland, building castles and founding noble families that would provide some future kings, such as Robert the Bruce, as well as founding a considerable number of the Scottish clans. King David I of Scotland, whose elder brother Alexander I had married Sybilla of Normandy, was instrumental in introducing Normans and Norman culture to Scotland, part of the process some scholars call the "Davidian Revolution". Having spent time at the court of Henry I of England (married to David's sister Maud of Scotland), and needing them to wrestle the kingdom from his nephew Máel Coluim mac Alaxandair, David had to reward many with lands. The process was continued under David's successors, most intensely of all under William the Lion. The Norman-derived feudal system was applied in varying degrees to most of Scotland.

== Mechanisms == The mechanisms by which healthy cells transform into cancer cells are described in detail elsewhere (see Cancer main page; Carcinogenesis main page). The precise molecular changes that result in sarcoma are not always known, but certain types of sarcomas are associated with particular genetic mutations. Examples include:
The analysis of the debris at the 10-megaton Ivy Mike nuclear test was a part of long-term project. One of the goals was studying the efficiency of production of transuranic elements in high-power nuclear explosions. The motive for these experiments was that synthesis of such elements from uranium requires multiple neutron capture. The probability of such events increases with the neutron flux, and nuclear explosions are the most powerful man-made neutron sources, providing densities of the order 1023 neutrons/cm2 within a microsecond, or about 1029 neutrons/(cm2·s). In comparison, the flux of HFIR is 5×1015 neutrons/(cm2·s). A dedicated laboratory was set up right at Enewetak Atoll for preliminary analysis of debris, as some isotopes could have decayed by the time the debris samples reached the mainland U.S. The laboratory was receiving samples for analysis as soon as possible, from airplanes equipped with paper filters which flew over the atoll after the tests. Whereas it was hoped to discover new chemical elements heavier than fermium, none of these were found even after a series of megaton explosions conducted between 1954 and 1956 at the atoll. The atmospheric results were supplemented by the underground test data accumulated in the 1960s at the Nevada Test Site, as it was hoped that powerful explosions in a confined space might give improved yields and heavier isotopes.
=== YxxCxxxF motif and GTS loop === The YxxCxxxF motif and GTS loop are two amino acid sites that are located in Domain 1 of eRF1. The YxxCxxxF motif is found in the amino acid residues 121-131, whereas the GTS loop is found in amnio acid residues 31-33. The YxxCxxxF is composed of three invariant amino acid residues: Tyrosine (Y), Cysteine (C), and Phenylalanine (F). These sites are structurally separated in the folded eRF1 protein, however their main functions are very similar. They are responsible for purine recognition in position 2 and 3 of the stop codon.
Biko and several other black African NUSAS members were frustrated when it organised parties in white dormitories, which black Africans were forbidden to enter. In July 1967, a NUSAS conference was held at Rhodes University in Grahamstown; after the students arrived, they found that dormitory accommodation had been arranged for the white and Indian delegates but not the black Africans, who were told that they could sleep in a local church. Biko and other black African delegates walked out of the conference in anger. Biko later related that this event forced him to rethink his belief in the multi-racial approach to political activism:

It is a synthetic peptide of 31 amino acids, built to resemble the natural incretin hormone GLP-1. Because of its size and composition it is handled analytically like other therapeutic peptides, using chromatographic and mass spectrometric methods rather than the techniques typical of small organic drugs.
The substitution at position 8 removes the site recognised by dipeptidyl peptidase-4, the enzyme that destroys the native hormone within minutes. The linked lipophilic chain then binds circulating albumin, which further limits access by degradative enzymes and reduces renal loss. Together these features lengthen the effective circulation time considerably.
It acts at the glucagon-like peptide-1 receptor, a G protein-coupled receptor that signals mainly through cyclic AMP. Activation is glucose dependent, meaning insulin release is stimulated more strongly when blood glucose is already elevated. Other tissues carrying the same receptor respond as well, which explains effects beyond glucose control.
Lower temperatures slow hydrolysis, oxidation and aggregation reactions that degrade the molecule over time. Lyophilised powder is more tolerant than solution, but both benefit from controlled conditions.
